چکیده

MicroRNAs (miRNAs) have recently been reported play a crucial role in some tumors. In order to investigate the association of miR-195 with hemangioma, we investigate the expression of miR-195 in hemangioma tissues and its role in biological behavior of XTPS-1 and HemEChemangioma cancer cell lines. Quantitative qPCR results showed miR-195 was significantly down-regulated in hemangioma tissues compared with normal tissues. Over-expression of miR-195 was established by transfecting miR-195 mimics into XTPS-1 and HemECs cells, and its effects on the biological behavior of hemangioma cancer were studied using transwell assay, MTT and flow cytometry. Ectopic expression of miR-195 promoted cell proliferation of hemangioma cancer cells, while loss-of-function assay showed the opposite effect. Results indicated up-regulation of miR-195 significantly inhibited cell proliferation, invasion and resulted in apoptosis of the hemangioma cancer cells. Moreover, we demonstrated that miR-195 down-regulated HIF-1a expression by directly targeting the HIF-1a 3’-untranslated region. Thus, our findings suggested that miR-195 acted as a new tumor suppressor by targeting the HIF-1a gene and inhibiting cell proliferation and invasion of hemangioma.

MicroRNA-195 induced apoptosis in hypoxic chondrocytes by targeting hypoxia-inducible factor 1 alpha.

OBJECTIVE The chondrocytes, the resident cells of cartilage, are maintained and take effects in the whole life upon chronic hypoxic exposure, which hypoxia-inducible factor 1 alpha (HIF-1α) play pivotal roles in response to. Dysregulation of some microRNA (miRNAs) have also been identified to be involved in hypoxia-related physiologic and pathophysiologic responses in some tissues or cell lines...

TFAP2A Regulates Nasopharyngeal Carcinoma Growth and Survival by Targeting HIF-1a Signaling Pathway

TFAP2A is a transcription factor that orchestrates a variety of cell processes, including cell growth and tissue differentiation. However, the regulation of TFAP2A in human nasopharyngeal carcinoma tumorigenesis and its precise mechanism of action remain largely unknown. In this study, we investigated the biologic role and clinical significance of TFAP2A in nasopharyngeal carcinoma growth and p...

HIF-1a promotes autophagic proteolysis of Dicer and enhances tumor metastasis

HIF-1a, one of the most extensively studied oncogenes, is activated by a variety of microenvironmental factors. The resulting biological effects are thought to depend on its transcriptional activity. The RNAse enzyme Dicer is frequently downregulated in human cancers, which has been functionally linked to enhanced metastatic properties; however, current knowledge of the upstream mechanisms regu...

MicroRNA-21 Down-regulates Rb1 Expression by Targeting PDCD4 in Retinoblastoma

Retinoblastoma (RB) is a children's ocular cancer caused by mutated retinoblastoma 1 (Rb1) gene on both alleles. Rb1 and other related genes could be regulated by microRNAs (miRNA) via complementarily pairing with their target sites. MicroRNA-21 (miR-21) possesses the oncogenic potential to target several tumor suppressor genes, including PDCD4, and regulates tumor progression and metastasis. H...

Dual Targeting of the Androgen Receptor and Hypoxia-Inducible Factor 1a Pathways Synergistically Inhibits Castration-Resistant Prostate Cancer Cells

Enzalutamide is a potent second-generation androgen receptor (AR) antagonist with activity in metastatic castrate-resistant prostate cancer (CRPC). Although enzalutamide is initially effective, disease progression inevitably ensues with the emergence of resistance. Intratumoral hypoxia is also associated with CRPC progression and treatment resistance. Given that both AR and hypoxia inducible fa...
